The mechanical basis of morphogenesis: I. Epithelial folding and invagination☆
[摘要] Wepresentamechanicalmodelforthemorphogeneticfoldingofembryonicepitheliabasedonhypothesizedmechanicalpropertiesofthecellularcytoskeleton.Inourmodelweconsiderasimplecuboidalepitheliumwhosecellsarejoinedattheirapicesbycircumferentialjunctions;tothesejunctionsareattachedcircumferentialarraysofmicrofilamentbundlesassembledintoa“pursestringâ€aroundthecellapex.Weassumethatthispursestringhasthefollowingproperty:ifitscircumferenceisincreasedbeyondacertainthreshold,anactivecontractionisinitiatedwhich“drawsthepurse-stringâ€andreducestheapicalcircumferenceofthecelltoanew,shorter,restinglength.Theremainderofthecellismodeledasavisoelasticbodyofconstantvolume.Clearlycontractioninonecellcouldstretchtheapicalcircumferencesofneighboringcellsand,ifthethresholdisexceeded,causethem“tofireâ€andcontract.Theobjectiveofthispaperistodemonstratethatourmodel,basedonthelocalbehaviorofindividualcells,generatesapropagatingcontractionwavewhichissufficienttoexplainthegloballycoherentmorphogeneticinfoldingofawidevarietyofembryonicepithelia.Representativecomputersimulations,basedonthemodel,arepresentedfortheinitialgastrulationmovementsofechinoderms,neuraltubeformationinurodeleamphibians,andventralfurrowformationinDrosophila.
