Fusion of chick embryo skeletal myoblasts: Interactions of prostaglandin E1, adenosine 3′:5′ monophosphate, and calcium influx☆
[摘要] Intheaccompanyingpaper(J.D.David,W.M.See,andC.-A.Higginbotham,1981,Develop.Biol.82,297–307)wedemonstratedthatanetcalciuminfluxintofusion-competentmyoblastsisarequisitestepinmembranefusion.R.J.Zalin(1974,Cell2,103–108;1977,Develop.Biol.59,241–248)hasshownthataprostaglandinE1(PGE1)-dependenttransientriseincAMPoccurs5–6hrpriortomyoblastfusion.Inthiscommunicationweshowthat(1)theincreaseinintracellularcAMPprecedes,and/orisindependentof,thecalciuminflux;(2)thecalciuminfluxiseitherdirectlyorindirectlydependentonPGE1activityaswellasPGE1synthesis;and(3)althoughthecAMPincreasemaybeessentialforfusion,itisnotsufficientintheabsenceofcalciuminflux.Ourexperimentsdefinefusioncompetency,ataminimum,as(1)theaccumulationofextracellularPGE1receptors;(2)theaccumulationofintracellularcAMPreceptors;and(3)theabilitytorespondtoacalciuminflux.
