Development of the flagellar rootlet during Naegleria flagellate differentiation☆
[摘要] Indirectimmunofluorescencestudiesofflagellatedifferentiationusingantiserumpreparedagainstthemajorrootlet(rhizoplast)proteinpermitustocompleteadescriptionofdevelopmentoftheflagellarapparatusofNaegleriagruberi.Thetimeandsequenceofrootletdevelopmenthavebeendescribedindifferentiatingcells,andrelatedtotheappearanceofbasalbodiesandflagellaandtotheassemblyofthemature,functionalflagellarapparatus.Developingrootlets,whichcannotbeidentifiedwithcertaintyuntiltheyare1.5-2μmlong,areseenintheearliesttransformingcells∼65minpostinitiation.Kineticsofrootletappearanceparallelthoseofbasalbodiesandflagella,withflagellaprecedingtherootletsbyabout5min(T50fordevelopingrootletsis76min).However,abrightlyfluorescingcondensateofrootletproteinappearsinthecytoplasmattheedgeofthenucleusat58min.Thispresumedinitiatingcenterforrootletassemblythuscondensesinthevicinityof,andvirtuallysimultaneouslywith,theflagellarbasalbodies.Rootletsattaintheirmaturelength(∼13μm)withinabout30minoftheirinitialappearance.Theythendisappearcompletelyfromcellsasflagellaarelostduringreversiontoamoebae.Subsequentlyrootletsarereassembledsimultaneouslywiththereappearanceofflagellaincellsundergoingasecondroundofflagellatedifferentiation.
