Development of rat neuromuscular junctions in organ culture☆
[摘要] Piecesofthoracicbodywall,includingintercostalmuscles,ribs,andthespinalcordwereexplantedfrom15to18-dayembryonicratsandmaintainedinorgancultureforupto6days.Duringthetimeinculturemusclefiberscontinuedtoincreaseinsize,andnervesproutsextendedalongthecenterofthemuscle.Whenmuscle-spinalcordexplantswereculturedat15daysgestation,thenumberofsynapticinputsperfiberincreasedwithtimeinculture.Subsequentlysynapseeliminationbeganwithatimecoursesimilartothatrecordedinvivo.In15-dayexplantsacetylcholinereceptorswereuniformlydistributedalongthefibersandfocalcholinesterase(ChE)wasnotdetected.Thecholinergicreceptorsstartedtoclusteratthemidregionofthefibersafter1dayexplantation,andChEwasdetectedinthefibersafter2daysinculture.ThecentralreceptorclusterswereassociatedwithChEandtheirformationwasdependentonthepresenceofnerveterminals.Weconcludethatneuromuscularcontactsdevelopinorganculturewithapatternandtimecoursesimilartothatofsynapesdevelopinginutero.
