Effects of the somatomedins and insulin on myoblast differentiation in vitro☆
[摘要] TheeffectsofinsulinandthesomatomedinsondifferentiationofratmyoblastswereinvestigatedinexperimentsoncellsclonedfromYaffe'sL6line.Incubationfor48hrwitheitherinsulinorTemin'smultiplicationstimulatingactivity(MSA),amemberofthesomatomedinfamily,causedadramaticincreaseinmyoblastfusion.Thisstimulationofdifferentiationisnotasimpleconsequenceoftheincreasedcelldensityresultingfromtheeffectsofthesehormonesonmyoblastproliferation,andtheincreaseinfusionisnotaneffectcommontoallmitogens(FGFinhibitstheprocess).Othersomatomedins(humansomatomedinCandinsulin-likegrowthfactorI),wereaseffectiveasMSAinstimulatingdifferentiation.Thesomatomedinswereactiveatconcentrationsintherangeoftheirlevelsinfetalblood,incontrasttoinsulin,whichwasinactiveatconcentrationsbelow10−7,M.Growthhormone(GH)hadnoeffectonmuscledifferentiation.Inserum-freemediumMM-1(inwhichmyoblastsmaintainapparentlynormalmorphologyandmetabolicactivity),theveryhighlevelsofinsulinrequiredtostimulatedifferentiationcouldbereplacedentirelybyphysiologicallevels(1.0μg/ml)ofMSA,furthersupportingourviewthatinsulinathighconcentrationsservesprimarilyasananalogueofthesomatomedinsinstimulatingthegrowthanddevelopmentofmusclecells.
