Activity and distribution of lysosomal enzymes during collagenous matrix-induced cartilage, bone, and bone marrow development
[摘要] Theappearanceofthelysosomalenzymesacidphosphatase,arylsulfatase,andβ-glucuronidasewasstudiedduringendochondralboneandbonemarrowformationinducedbyimplantationofdemineralizedbonematrix.Theactivitiesofacidphosphataseandβ-glucuronidasegraduallyincreasedfromthestageofmesenchymalcellproliferationonDay3onwardtoreachapeakonDay13,duringmaximalboneremodeling.However,arysulfataseactivityexhibitedasharpincreaseonDay9,associatedwiththeonsetofcartilagehypertrophyandchondrolysis.ThepeakofarylsulfataseactivitywasalsoattainedonDay13.TheactivitiesofallthreeenzymesdeclinedonDay15butacidphosphataseagainexhibitedanincreaseduringhematopoieticbonemarrowdifferentiationonDays19–21.Histochemicalandultrastructuralstudiesrevealedintenselysosomalenzymeactivityinmacrophage-likecellsonDay7andthereafter.Duringchondrolysisandboneremodeling,thesecellswerepresentinaperivascularlocation.Osteoclastsalsoexhibitedstrongreactivityforthelysosomalenzymes.Duetoitscharacteristictemporalappearanceduringdevelopmentofendochondralbone,arysulfatasemaybeusedasamarkerenzymeforchondrolysisandboneresorption.
