Studies on glycosaminoglycans of regenerating rabbit ear cartilage☆
[摘要] Cartilageregenerationintheadultrabbitearwasexaminedwithrespecttoglycosaminoglycan(GAG)synthesisatvariousstagesoftheregenerationprocess.Increasedhyaluronicacidandchondroitinsulfatesynthesiswasfirstseen31daysafterwounding,whenametachromaticcartilagematrixcouldbedistinguishedfromblastemalcells.Analysisofcartilageandtheoverlyingskinseparatelyshowedthat90%ofthelabeledchondroitinsulfatewasfoundinthecartilagebeingregenerated.DEAE-cellulosechromatographyofGAGpreparationsfrom35-dayregeneratingcartilagesshowedhyaluronicacidandchondroitinsulfatepeakselutinginthesamepositionasthoseisolatedfromnormalcartilages.TheidentityofthehyaluronicacidandchondroitinsulfatepeakswasconfirmedbytheirsusceptibilitytoStreptomyceshyaluronidaseandchondroitinaseABC,respectively.Althoughthedegreeofsulfationinnormalandregeneratedcartilageswassimilar,theratioofchondroitin6-sulfatetochondroitin4-sulfatewasincreasedinregeneratedcartilages.GAGpreparationsfromunlabeledcartilagesweredigestedwithchondroitinaseABCandthedisaccharidedigestiveproductswereidentifiedandquantitiated.NormalcartilagehadaΔDi-6SΔDi-4Sratioof0.27;thesameratiofortheregeneratedcartilagewas1.58.
