The developmental potential of synchronized amphibian cell nuclei☆
[摘要] SinglecellnucleifromsynchronizedRanapipienscellculturesweretransplantedintoenucleatedRanapipienseggstodeterminewhetherthereisarelationshipbetweenthephaseofthecellcyclefromwhichadonornucleusistransplantedanditsabilitytopromotenormalembryogenesis.SynchronizedpopulationsofICR2Acells,astablehaploidcellline,wereobtainedbymitoticselection.NucleiweretransplantedattimesafterselectioncorrespondingtoG1,mid-S,lateS,andG2.Nucleartransplantswerealsomadefromunsynchronizedpopulationsandhaploidblastulae.TheresultsoftheseexperimentsindicatedthatdonornucleifromG1cellshavearestrictedcapacitytosupportdevelopmentbeyondtheblastulastage.Nosignificantdifferencescouldbedetectedinthedevelopmentalcompetenceofnucleiderivedfrommid-S,lateS,G2,andunsynchronizednuclei.Dataonthefrequencyofchromosomalabnormalitiesinnucleartransplantembryos,nuclearswelling,andthymidineincorporationbytransplantednucleialsosuggestthatG1nucleihaveadecreasedabilitytoreplicatetheirchromosomesineggcytoplasm.Itisconcludedthatdonorcellcyclephasedoeshaveaneffectonnucleartransplantationsuccess.
