In vivo and in vitro differentiation of neurons and astrocytes in the rat embryo: Immunofluorescence study with neurofilament and glial filament antisera☆
[摘要] Antiseraraisedagainstneurofilament(NF)peptidesandglialfibrillaryacidicprotein(GFA)(subunitofglialfilaments)havebeenusedtoidentifyneuronsandastrocytesinordertostudytheirdevelopmentanddifferentiationinratembryo.InvivoobservationsshowedthatNF-positivecellsfirstappearedin12-day-oldembryos,whereasGFA-positivecellsappearedinbrainandspinalcordonthe18thday.InvitroobservationsshowedthatNF-positivecellscouldbeobtainedonlyinculturesfrom12-dayembryosonwards.Thefurtherdifferentiationofneuronsinvolvedneuriteelongation,aggregationofcellbodiestoformislets,andemergenceofverybrightlystainingprominentneuronswithlargecellbodiesandlongneuriteswhichtookpartincomplicatepatternformation.GFA-positivecellsappearedinvitroonthe16thdayandtheycouldbeobservedeveninculturesobtainedfrom10-day-oldembryos.Asthecultureaged,theGFAstainingbecamehighlyfibrillary.Therewasnophysicalinteractionbetweenneuronalandglialprocesses.
