Chick embryo intestine in culture: Superinduction of alkaline phosphatase and maltase activities by actinomycin D☆
[摘要] Duodenumofthe14-daychickembryoexplantedtosyntheticmediumwithoutserumorhormonescarriesonstructuralandbiochemicaldifferentiationatafasterratethaninsitu.Suchdifferentiationisinhibitedby7μMcycloheximideor1.6μMactinomycinDwhenpresentthroughoutthe48-hrcultureperiod.IfactinomycinDisaddedafter8or12hr,theincreaseofalkalinephosphataseandmaltaseactivitiesthatoccursincontrolculturesisaccentuatedratherthaninhibited,sothatactivitiesattheendof48hraresignificantlygreaterthanthoseattainedintheabsenceoftheinhibitor.IfactinomycinDisremovedafter2–4hr,alkalinephosphataseandmaltasereachevenhigherlevelsattheendof48hrthanwithdelayedaddition.Explantsof18-dayduodenumarebycontrastinsensitivetoactinomycinD,attainingsimilarlevelsofactivitiesofbothenzymesinthepresenceorabsenceoftheinhibitorduringtheentirecultureperiod.Theseresultsmaybeinterpretedintermsofanactinomycin-sensitiverepressorthatnormallydeclinesduringlateembryoniclife,andfailstobemaintainedinvitro.
