Regulation and secretion of early developmentally controlled enzymes during axenic growth in Dictyostelium discoideum☆
[摘要] Thefourearliestdevelopmentallycontrolledenzymesinthecellularslimemold,Dictyosteliumdiscoideum,accumulateduringaxenicgrowthinrichmedia.WehaveshownthatatlowcelltitersthespecificactivitiesofN-acetylglucosaminidase,α-mannosidase,leucineaminopeptidase,andalaninetransaminaseareeachatveryloworvegetativelevelscomparabletoamoebaewhichhavebeengrownonbacteriaasthefoodsource.Duringtheexponentialphaseofgrowthallfourenzymesaccumulatedramaticallyreachingcellularspecificactivitiesatleastashighasduringdevelopment.Themagnitudeofthisaccumulationisinfluencedbyalterationsinthegrowthmedium.Wesuggestthattheseresults,combinedwiththoseofpriorinvestigations,indicatethatarestrictedsegmentofearlydevelopmentisinitiatedduringaxenicgrowth.Thismeansthatgrowthandearlydevelopmentarenotmutuallyexclusiveeventsinthisorganism.Thesecretionoflysosomalenzymesisalsoaffectedbythecompositionofthegrowthmedia.Inallmedia,includinggrowthinbacterialsuspensions,lysosomalenzymesaresecretedinsignificantquantities.Thereisacorrespondenceintheeffectsofmediacompositiononthesecretionoftheseenzymesandontheregulationofdevelopmentallycontrolledenzymesduringaxenicgrowth.Thesecretionoflysosomalenzymesthatarenotdevelopmentallyregulatedisaffectedinthesemedia,suggestingthattheregulationandsecretionoftheseenzymesareunderseparatecontrol.Itisclearthatstudiesoftheregulationoflysosomalenzymesinthisorganismmusttakeintoaccountthesecretionoftheenzymesaswellastheircellularspecificactivitiestoproperlyreflectlevelsofgeneexpression.
