Cell-cell adhesion in the embryonic chick: Partial purification of liver adhesion molecules from liver membranes☆
[摘要] Polyspecificxenoantiseraspecificforembryonicchicklivercellshavebeenusedtodevelopanassayforthedetectionofembryonicchickliveradhesionmolecules(LAM).Theneutralizationoftheantiaggregationimmuneseraandsubsequentsemiquantitativeassessmentoftheremainingactivityisthebasisforthisassay.EmbryonicchicklivermembraneproteinshavebeensolubilizedusinganumberofdetergentsandtheLAMactivityhasbeenpartiallypurifiedusingsucrose-gradientsedimentation,isoelectricfocusing,gelfiltration,affinitychromatography,andpreparativepolyacrylamidegelelectrophoresisprocedures.TheisolatedLAMappearstobeanintrinsic,trypsin-resistant,membraneglycoproteinofapproximately65,000daltons.The65KpreparationofLAMisaneffectiveimmunogeningeneratinganaggregationblockingxenoantiserumspecificforembryonicchicklivercells.PurificationofLAMtohomogeneityhasbeenhamperedbyanapparentlabilitybyself-associationorlossbyadsorptionofthemorepurifiedLAMantigen(s).
