Membrane properties of cultured rat sympathetic neurons: Morphological studies of adrenergic and cholinergic differentiation☆
[摘要] Dissociatedneuronsfromthenewbornratsuperiorcervicalganglionweregrownunderconditionswhichleadtoeitheradrenergicorcholinergicdifferentiation.Lectinsandtoxinswereusedtodetectdifferencesinthecellmembraneassociatedwithtransmitterstatus,ageoftheneurons,orlocationontheneurons.Theseligandsweremadevisibleinthelightorelectronmicroscopebycouplingtorhodamineorcolloidalgold.ThedensityofbindingsitesforconcanavalinA(ConA),ricin(RCA60),andwheatgermagglutinin(WGA)increasedwithageincultureonbothadrenergicandcholinergiccells.Soybeanagglutinin(SBA)bindingincreasedaboutthreefoldonadrenergicaxons,butfailedtoincreaseonneuronsinducedtobecomecholinergicbymediumconditionedbyratheartcells(CM).TheeffectofCMonSBAbindingparalleledpreviouslydescribedeffectsofCMontransmitterproduction;theCMbindingpatterndevelopedslowlyandwasnotreadilyreversible.MatureadrenergicneuronsalsoappearedtobindmoreWGAthanneuronsinCMcultures.Tetanustoxingoldbindingwasuniform,butlow,onaxonsofadrenergicandcholinergicneuronsatallages.Incontrast,choleratoxinbindingdecreasedwithageonadrenergicaxons.BindingsitesforSBAandtetanustoxinwerefoundtobelessnumerousonthecellbodysurfacethanontheaxonalsurface.ThusgrowthinCMinducesfundamentalchangesinthephenotypeofdevelopingsympatheticneuronsinvolvingthecellmembraneaswellastransmitterchoice.Differencesalsoappearwithmaturationandbetweenaxonalandsomaticcellsurfacemembranes.
