Modulation of cell-to-cell communication in the cumulus-oocyte complex and the regulation of oocyte maturation by LH☆
[摘要] Priortoovulation,cell-to-cellcommunicationbetweentheoocyteandthecellsofthecumulusoophorusisterminated(N.B.Gilula,M.L.Epstein,andW.H.Beers,1978,J.CellBiol.78,58–75;R.M.Moor,M.W.Smith,andR.M.C.Dawson,1980,Exp.CellRes.126,15–29).InthispaperwereportthatLH,Bt2-cAMP,andthecyclicnucleotidephosphodiesteraseinhibitor,3-isobutyl-1-methylxanthine,areallcapableofinterruptingcommunicationinvitroinratfollicle-enclosedcumulus-oocytecomplexes.Moreover,thebreakdownofcommunicationappearstobecloselycorrelatedwiththeabilityofhyaluronidasetodispersethecumuluscellmass.Thisobservationallowssimplescreeningoftheeffectsofvariousagentsoncumulus-oocytecommunication.Theinvitrosystememployedinthisstudyhasalsobeenusedtoinvestigatetherelationshipbetweencommunicationandoocytematuration.Thefindingspresentedindicatethattheinterruptionofcommunicationbetweenthecumulusandtheoocyteleadstoreliefofmeioticarrest,andareconsistentwiththepossibilitythatcAMP,transmittedfromthecumulustotheoocyte,maybetheinhibitorofoocytematurationinvivo.
