The effect of cytochalasin B and D on the fertilization of sea urchins☆
[摘要] Twoelectricaleventsoccuracrosstheplasmamembraneoftheseaurchineggduringtheinteractionwiththefertilizingspermatozoon;thefirst,astep-likedepolarizationpecedesthefertilizationpotential(FP)by11secat20°C.CytochalasinsBandDaltertheseeventsinseveralwaysdependingupondoseandtimeofexposureofthegametes.Whenboththeeggandthespermatozoonareexposedtothesemicrofilament-activedrugs,theformerforminuteswhilstthelatterintheorderofseconds,thedelaybetweenthetwoelectricaleventsisincreasedtwofold.Wesuggestthatamicrofilament-involvedprocessisoccurringduringthisdelayperiodwhich,althoughitselfnotalteringtheelectricalpropertiesoftheplasmamembrane,determinesthetimeofinitiationoftheFP.PretreatingspermatozoawithhigherdosesofeitherCBorCD,inhibitsfertilizationcompletelyorlowersthesuccessfulcollisionrate(α);wesuggestthatthetwodrugslowerαbyaffectingtheacrosomalregionofthespermatozoon.AlthoughbothdrugsincreasethedelayperiodtothesamedegreeCDhasfewersecondaryeffectsthanCB.
