Sorting out of coaggregated brain cell types mediated by specific cell recognition factors in vitro: II. Kinetic and metabolic parameters☆
[摘要] ThetimecourseofcerebrumcellassociationpromotedbyCRF(acerebrum-specificcellrecognitionfactorrecoveredfromsupernatantmediaofembryoniccerebrumcellcultures)wasdeterminedwithrespectto(a)theincreaseinsizeofaggregatesformedfromdissociatedcerebrumcellsand(b)thesortingoutofcerebrumcellsfromheterotypiccellswithincoaggregates.Botheventswereinitiatedapproximately5hrafterthesingle-cellsuspensionswereplacedinculture.WhilethesizeofaggregatesformedbycerebrumcellsinresponsetoCRFcontinuedtoincreasethroughouttheremainderofthe24-hrcultureperiod,thesortingoutofcerebrumcellswithincerebrum-tectumcellcoaggregateswascompletedbetweenthefifthandsixthhourofculture.IntheabsenceofexogenouslyadministeredCRF,approximately3dayswasrequiredforcerebrumandtectumcellstosortoutwithincoaggregatecultures.Studiesatlowtemperatureorinthepresenceofsodiumazideand2-deoxyglucoseindicatedthatcellsortingwasanenergy-dependentprocess.Althoughproteinsynthesiswasrequiredduringtheinitial1–2hrofcultureforcoaggregationtoproceed,thecellsortingprocessmediatedbyCRFdidnotrequireconcomitantproteinsynthesis.Theparticipationofmicrotubuleandmicrofilamentsystemsinthecellsortingeventswasexaminedbytheuseofthefibrillarinhibitors,colchicineandcytochalasinB.Thesestudiesdemonstratedthatsortingoutoccurredinthepresenceofcolchicine,butwashighlysensitivetocytochalasinB.
