Membrane-bound and free polysomes in Dictyostelium discoideum: I. Isolation and developmental effects on size and distribution☆
[摘要] Usingasimpleone-stepprocedureundegradedmembrane-boundpolysomesrelativelyuncontaminatedwithfreepolysomeswerepurifiedfromthecellularslimemold,Dictyosteliumdiscoideum.Inaxenicallygrownvegetativecells17–18%ofthetotalribosomesareassociatedwithmembranes.Thisvaluedropsslightlyduringtheaggregationstageofdevelopment,increasestovegetativelevelsinpseudoplasmodia,andagaindropsslightlyduringfruitingbodyconstruction.Within15minafterdevelopmentisinitiatedbytheremovalofnutrients,boththetotalandmembrane-boundpolysomesdecreaseinsizefromanaverageof9–11ribosomespermRNAtoanaverageof7or8ribosomespermRNA.Thepercentageoftotalribosomesfoundasmonosomesincreasesfrom10to55%.Bothtotalandmembrane-boundpolysomesremainatthisreducedsizethroughoutdevelopment.
