Oriented extracellular channels and axonal guidance in the embryonic chick retina☆
[摘要] Transmissionelectronmicroscopyandserialreconstructionoflumsectionswereusedtodeterminewhetheralignedextracellularchannelsprecedetheoutgrowthofopticfibersintheembryonicchickretina.Atstage16,justpriortothemigrationofopticaxonstowardtheopticstalk,extracellularspacesboundedbyneuroepithelialcellprocesses,inthesuperficial(vitread)regionoftheretina,werealignedtowardtheopticstalk.Theopticaxonssubsequentlyenteredandgrewwithinthesespaces.Afterformationoftheganglioncellfiberlayer(GCFL),thegrowthconesofnewopticaxonsenteredthemostvitreadportionofthatlayer.Hypertonicfixativescausedshrinkageofcellprocesses,resultinginintercellularseparation.However,growthconefilopodiaretainedclosecontactswithneighboringglialcellendfeetandwithopticaxonsinthesesolutions.Thissuggeststhatgrowthconesmaybeadherenttothesestructures.Transmissionelectronmicroscopyinconjunctionwiththeuseofhypertonicsolutionsmaybecomeausefultechniqueforassayingintercellularadhesivity.
