Erasure of the memory response in MEL cells☆
[摘要] WhenMELcellsarereexposedtoDMSOafteraninterruptionininducertreatment,theycaninitiatecommitmenttodifferentiationwithoutthelagperiodobservedaftertheprimaryexposuretoinducer.Thispropertyisknownasmemory.Herewehaveemployedmetabolicinhibitorstoanalyzethebasisofthememoryresponse.Treatmentofcellswithcycloheximideorcordycepinduringtheinducerwithdrawalperiodcausesmemoryerasure.CellsmustrecapitulateanentirelagperioduponreexposuretoDMSO.Thememoryresponseismaintained,however,ifcellsaretreatedwithmetabolicinhibitorsinthepresenceofDMSO.OurresultssuggestthatthecapacityofMELcellsformemoryrequiresthesynthesisofcellcomponentswhicharenormallystableintheabsenceofDMSO.Experimentsinvolvingreciprocalshiftsbetweentwodifferentinhibitorshavebeenperformed.Evidenceispresentedthattheprocessleadingtotheinitiationofcommitmentiscomposedofatleastthreecomponentsactinginsequence.
