Ligatin from embryonic chick neural retina inhibits retinal cell adhesion☆
[摘要] Ligatin,afilamentouscell-surfaceproteinpurifiedfromembryonicchickneuralretina,hasbeenfoundtoinhibitthereassociationofdissociatedretinalcells.Thisinhibitionwasdemonstratedusingtwomethods,asinglecelldisappearanceassayandanimprovedmonolayercollectionassayutilizingmicrotiterplates.Monomericligatinatapproximately20μg/mlinhibitedratesofadhesion,butpolymericligatinandtrypticfragmentsofligatinwereineffective.Ligatin'sinhibitoryeffectissuggestedtobemediatedthroughbindingtoretinalcellsurfacessincepreincubationofdissociatedretinalcellswithmonomericligatininhibitedthecells'adhesivenessandremovedtheinhibitoryactivityfromtheculturemedia.Ligatinhomologuespreparedfrommammaliantissueswereineffectiveininhibitingretinalcelladhesion,suggestingatissueand/orspeciesspecificity.SimilaritiesinphysicochemicalandbiologicalpropertiessuggestthatligatinmaybetheinhibitorofadhesionpreviouslydescribedbyMerrelletal.[Merrell,R.,Gottlieb,D.I.,andGlaser,L.(1975).J.Biol.Chem.,250,4825].
