Juvenile hormone-stimulated polyploidy in adult locust fat body☆
[摘要] TherelativeDNAcontentsofadultfatbodynucleiofLocustamigratoriahavebeenassayedbyscanningmicrospectrophotometryafterFeulgenstaining,spermatidandbrainnucleibeingusedashaploidanddiploidreferencestandards.Inthefatbody,severalDNAclasseswerefound,themeanploidyincreasingduringtheperiodofreproductivematuration.Theextinctionfrequencypeaksfallatvaluesthatarelessthandoublingsofthediploidvalue,whichsuggestsincompletereplicationofthegenome,althoughthisinterpretationrequiresconfirmationbyindependenttechniques.OnDay1thefemaleandmalefatbodynucleiarechieflytetraploids.Elevationtooctaploidyorhigher,especiallyinthefemaleisevidentbyDay15.Allatectomy,removingthesourceofJH,preventstheincreaseinploidy,andadministrationoftheJHmimic,ZR-515,restoresitinbothsexes.Incorporationof[3H]thymidineinfemalefatbodyshowsabroadpeak,maximalatDay6.Afterallatectomy,incorporationof[3H]thymidineisstronglystimulatedbyZR-515inbothsexes.ItissuggestedthattheJH-dependentpolyploidyinfemalefatbodypermitsacceleratedproductionofmRNAforvitellogeninandpossiblyotherproteins.ThemalefatbodyrespondstoJHbyDNAreplicationbutnotbyexpressionofthevitellogeningene.
