Developmental expression in central and peripheral nervous system of oligodendrocyte cell surface antigens (O antigens) recognized by monoclonal antibodies
[摘要] FourantigensdesignatedO1,O2,O3,andO4havebeendetectedbymousemonoclonalantibodies.Theyareexpressedonthecellsurfacesofoligodendrocytesinthecentralnervoussystem(seetheprecedingarticle,I.SommerandM.Schachner(1981,Develop.Biol.83,311–327)).Oantigensaredetectableonfreshlytrypsinizedviablesinglecellsuspensionsfromearlypostnatalcerebellumbyindirectimmunofluorescenceprocedures.WhereasantigensO1andO2arenotdetectableatbirth,O3andO4antigensarerecognizedatthisage;atpostnatalDay7allOantigensaredetectable.InmonolayerculturesoffetalponsandcerebellumexpressionofantigensO3andO4precedesthedetectabilityofgalactocerebrosideandantigensO1andO2.Inculturesof7-day-oldmousecerebellumO1andalmostallO2antigen-positivecellsarealsogalactocerebroside-positive,whereasO3andO4antigensareadditionallypresentonmoresimpleorimmature-lookinggalactocerebroside-negativecells.TheseO3andO4antigen-positive,butgalactocerebroside-negativecellsareneitherpositiveforGFAprotein,tetanustoxinreceptors,norfibronectin.After7daysinculturemoreO3andO4antigen-positivecellsaregalactocerebroside-positivethanafter2days.Intheperipheralnervoussystem,in2to7-day-oldculturesofneonataldorsalrootganglioncells,O1andO2antigensarehardlydetectableunderthecultureconditionsused,whereasantigensO3andO4arefoundonsomebutnotallSchwanncells.After7daysinvitrofewerSchwanncellsarepositiveforO3andO4antigensthanat2days.AntigenO4isfoundadditionallyonthesurfaceofdorsalrootganglionneurons,particularlyafterprolongedcultureperiods.
