Patterns of temperature sensitivity in Contrabithorax/Ultrabithorax heterozygotes of Drosophila☆
[摘要] Contrabithorax(Cbx)isadominanthomeoticmutantofDrosophilawhichtransformswingstohalteres,whileUltrabithorax(Ubx)isadominantmutantwhichtransformshalterestowings.ThereforeCbxUbxfliescarrydominanthomeoticmutantsengagedinopposingtransformations.ThisarticlereportsthatCbxTM2Ubx130istemperaturesensitive.At29°C,fliesexpressstrongCbxtransformationofwings,andminorUbxtransformationofhalteres.Larvaeshiftedto17°Cpriorto72hrexpressstrongUbxtransformationofhalterestowardwings,andslightCbxtransformationofwings.Seventeen-degreetemperature-pulseexperimentsshowthattheCbxTM2Ubx130systemistemperaturesensitivecontinuouslyduringembryonicandlarvallife.ExpressionoftheCbxtransformationinleftandrightwingsishighlycorrelatedinallconditionsstudied,asisexpressionoftheUbxtransformationinleftandrighthalteres,buttheCbxtransformationinwingsandUbxtransformationinhalterescanbenegativelycorrelatedoruncorrelated.ThetemperaturesensitivityinCbxTM2Ubx130isnotfoundinCbxUbx61D,andUbxisonlyweaklyexpressedinSbTM2Ubx130flies.TheseresultsshowthatCbxintranstoUbxcanenhanceUbxexpression,althoughCbxalsocausesanopposingtransformationtoUbx;thattheCbxUbxsystemactsinboththemesothoraxandmetathoraxtomodulatetheirphenotypes;andthatbothtransformationsarebroadlytemperaturesensitivethroughembryonicandlarvallife.ThissuggeststhattheCbxTM2Ubx130systemfunctionscontinuouslyduringembryonicandlarvaldevelopmenttomaintainmesothoracicandmetathoraciccommitments.TheresultsareinterpretedintermsofaCbxUbxfeedbackloopwhichmaintainsthemesothoraxinastateoflowCbxUbxactivity,andmetathoraxinastateofhighCbxUbxactivity.
