Ontogeny of maternal and newly transcribed mRNA analyzed by in situ hybridization during development of Caenorhabditis elegans
[摘要] Inthepresentinvestigationwehaveexaminedthetiterofpoly(A)insquashespreparedfromoocytesandvariouslystagedembryosofthenematodeCaenorhabditiselegansvar.Bristol(N2)byinsituhybridizationwitha[3H]poly(U)probe.AsshownbycontrolexperimentsinwhichsquashesweretreatedwithdilutealkaliandRNasepriortoinsituhybridization,thisprobeinteractsspecificallywithpoly(A)sequenceswhicharepresumablypresentinpoly(A)+RNA.Usingthismethodandsaturatingconcentrationsoftheprobe,itwasshownthatisolatedoocytesandembryosuptothe125-cellstageexhibitedsubstantiallevelsofpoly(A)intheircytoplasmsbutnodetectableamountsofthissequencewithintheirnuclei.Nuclearpoly(A)wasfirstdetectedatthe90-cellstageandafterwardincreasedatalinearratethroughthe550-cellstage.Thetiteroftotalembryonicpoly(A)alsoincreasedatalinearrateuptolarvalhatching.Theresultssuggestthatthemajortranscriptionaleffortforpoly(A)+RNAbeginsatthe90-to125-cellstageofC.elegansembryogenesisandthatthepoly(A)+RNApresentinthecytoplasmpriortothistimeisprimarilyofmaternalorigin.
