Effects of α-amanitin on programming of mouse blastocyst development
[摘要] Wehaveinvestigatedtheeffectofα-amanitintreatmentofmouseblastocystsontheirfurtherdevelopment.Twenty-four-hourtreatmentswithα-amanitinat1μg/mldonotinterferewithembryosurvivalorgrossmorphologicaldevelopmentoverthenextfewdaysalthoughtherateofsynthesisofpolyA-containingRNAisdramaticallyreducedduringexposuretotheantimetabolite.Effectsuponratesofproteinsynthesisarenotobservedinitially,butincorporationof(35S)methionineintoacid-insolublematerialisultimatelyinhibitedbyapproximately50%inallthetreatedsamples.Developmentalmarkersdonotallrespondinthesamewaytotheα-amanitintreatmentsused.Whereaslevelsofβ-glucuronidaseandplasminogenactivatorarevirtuallyunaffectedbytheantimetabolite,α-amanitintreatmentinterfereswithΔ5,3β-hydroxysteroiddehydrogenaseactivityintrophoblastcellsandformationofabilayeredinnercellmass.Theeffectsuponthelattermarkersdifferquantitativelywiththevariousexposureperiodstoα-amanitin.Onthebasisoftheseobservations,weproposethatmRNAsforatleastsomedevelopmentalmarkersinmouseblastocystsaresynthesizedlongbeforetheyaretranslated.
