Temporal changes in lectin binding of peri-implantation mouse blastocysts☆
[摘要] Mouseblastocystswereexposedtoaseriesofferritin-conjugatedlectinsduringDay5(preadhesive)andDay6(adhesive;collectedDay5,24hrinvitro)ofembryogenesistodeterminewhethertherewereanychangesinlectinbindingcharacteristicsthatcoincidedwiththeacquisitionofadhesiveness.Afterexposuretolectin,theblastocystswereprocessedforelectronmicroscopyandlectinbindingsitesweredeterminedbyvisualizationofferritinparticleswiththeelectronmicroscope.NobindingsiteswereobservedforeitherDolichosbiflorusagglutininorsoybeanagglutininonblastocystsfromeitherstageexamined.BindingsitesforUlexeuropaeusagglutinin,ConA,andwheatgermagglutininwereseenonblastocystsfrombothstageswithoutapparentincreaseorreductioninbindingsitesfromeitherstage.Ricinuscommunisagglutinin-I(RCA-I)boundheavilytothesurfaceofDay5blastocystsanddidnotbindatallto312Day6blastocystsanddidbind,thoughwithapparentdiminution,to912Day6blastocysts,ascomparedwiththebindingobservedonDay5blastocysts.Peanutagglutinin(PNA)didnotbindatalltoDay5blastocystsbutdidbindheavilytothesurfaceofDay6blastocysts.BothRCA-IandPNAboundtothesurfaceofembryosduringDay5ofdelayedimplantation,thusindicatingthatneithertheappearanceofPNAbindingsitesonDay6blastocystsnortheapparentreductionofRCA-IbindingsitesonDay6blastocystscouldbesolelyimplicatedintheacquisitionofadhesiveness.PNAbindingsiteswereabolishedfromthesurfaceofDay6blastocystsbytreatmentwithPronase,indicatingthatthePNAbindingmoleculewasassociatedwithaglycoproteinratherthanaglycolipid.
