Astrocytes, ependymal cells, and oligodendrocytes develop on schedule in dissociated cell cultures of embryonic rat brain
[摘要] Wehaveusedcell-type-specificmarkerstostudythedevelopmentofneuronesandglialcellsindissociatedcellculturesof10-dayembryonicratbrain.Tetanus-toxin-bindingneuronesandphagocyticmacrophageswithsurfacereceptorsforIgGwerepresentfromthebeginningofsuchcultures.GFAP+astrocytesdevelopedafter5–6days,ependymalcellswithbeatingciliaafter7–8days,andgalactocerebroside+oligodendrocytesafter13–14daysinculture.Whencultureswerepreparedfrom13-dayembryonicbrain,theseglialcellsdeveloped3daysearlierthantheydidinculturesof10-dayembryonicbrain.Whenfreshlyisolatedcellsuspensionsfrombrainsofembryosatvariousageswerestudied,wefoundthatthedevelopmentofspecificneuralcellsinvivopreciselyparallelledtheirdevelopmentinvitro.Thusglialcellsdeveloprightonscheduleindissociatedcellculturessuggestingthatbiologicalclocksaremoreimportantthanpositionalinformationingliogenesisafter10daysgestation.Wedemonstratethatratneuralantigen-2(Ran-2)whichisdefinedbyamonoclonalantibodyisexpressedonastrocyteprecursors.
