Quantitation of the accumulation of histone messenger RNA during oogenesis in Xenopus leavis☆
[摘要] TheaccumulationofmessengerRNAcodingforhistoneH3inoogenesisofXenopuslaeviswasstudiedbyquantitativehybridizationtechniques,usingaclonedgenomicDNAfragmentasaprobe.ThisprobewasisolatedfromclonedXenopushistoneDNAandcontainsmostoftheH3codingsequences.HistoneH3mRNAaccumulationwasfoundtobecompletedbeforethemaximumlampbrushstage.HybridizationofRNAblotswithDNAprobescontaininggenesforhistonesH2A,H2B,andH4suggeststhesameaccumulationpatternforthemRNAscodingforthesehistonesasforhistoneH3mRNA.TheamountofH3mRNAinthematureoocytewasestablishedtobe130±68pg,i.e.,about5×108copies.
