The spatial distribution of hyaluronic acid and mesenchymal condensation in the embryonic chick wing☆
[摘要] Thedevelopmentandlocalizationofthemesenchymalextracellularmatrix(ECM)intheearlyembryonicchickwingwasstudiedbyTEMusingafixativewhichenhancedthepreservationofglycosaminoglycans,especiallyhyaluronicacid(HA)(SingleyandSolursh,Histochemistry65,93–102,1980).Fromstage19tostage24ofHamburgerandHamiltontherewasaprogressiveincreaseinthedensityofECMinthemuscleandlooseconnectivetissueformingperipheralregion.Bycomparison,thedensityofECMremainedverylowintheprechondrogeniccore.ThemajorcomponentoftheECMresponsiblefortheobservedincreaseindensityappearedtobeHA,basedonitsmophologyandlabilitytoStreptomyceshyaluronidase(S.Hase).Atstage26theconcentrationoftheHA-likematrixcomponentwasgreatlyreducedinthemyogenicareasand,toalesserdegree,inareasoflooseconnectivetissueformation.ThedistributionofS.Hase-sensitivegrainsoverextracellularareasinautoradiogramsof[3H]glucosamine-labeledwingsatstage24paralleledthedistributionofECM.InjectionofS.HaseintowingbudsinovoresultedinadrasticreductionintheamountofputativeHA-richmatrixandamarkedreductionofintercellularspaces(ICS).Experimentallyinducedhypotensioninembryospriortofixationresultedinasimilar,butmuchlessdrastic,reductionofICSinthelimbperipheryandamarkedincreaseinthedensityoftheECM.ThespatialorganizationoftheECMintheearlylimbanditssensitivitytohypotensionareconsideredinrelationtotheclassicalphenomononofformationofmesenchymalcondensations.
