Accumulation in embryogenesis of five mRNAs enriched in the ectoderm of the sea urchin pluteus☆
[摘要] FivecDNAclonesrepresentingmRNAsenrichedintheectodermoftheseaurchinpluteushavebeenisolatedfromalibraryof1000pluteuscDNAclones.WehaveusedRNAgelblotstoshowthattheRNAsarepresentatlevels3to20timeshigherintheectodermthanintheendoderm/mesoderm.BasedongenomicDNAgelblotsthefivedifferentmRNAswereplacedintofourgroups.ThreeofthemRNAsbelongtoseparategroups,andtheremainingtwomRNAsaremembersofafourthgroup.TheaccumulationofthemRNAsduringembryogenesiswasmeasuredusingtotalcellularRNAfromunfertilizedeggs,16-cell,mesenchymeblastula,mid-gastrula,andpluteus-stageembryos.AllofthemRNAsarepresentatveryloworundetectablelevelsintheunfertilizedeggand16-cellembryo.TheamountsthenincreasesharplyandreachamaximumatblastulaorgastruladependingontheparticularRNA,andthendeclinesignificantlybypluteus.Inmostofthecases,RNAlevelschangeover100-foldinlessthan48hrofdevelopment.Thisaccumulationisduetotranscriptionduringembryogenesis.ThemRNAsarefoundonpolyribosomesatblastulaandpluteusstages,butthenatureoftheproteinstheyencodeisunknown.
