Neural differentiation following culture of embryonal carcinoma cells in a serum-free defined medium
[摘要] TheembryonalcarcinomalineC17-S1clone1003ismultipotentialinvivo.Whenthecellsaregrowninvitroinserum-containingmediummostofthemremainundifferentiated,whileafewdifferentiateintoauniquemorphologictypeofepithelioidcell.If1003cellsarepassagedintoadefinedmediumcontaininginsulin,transferrin,selenium,andfibronectintheygrowforsixtoeightgenerationsatthesamerateasinserum-containingmedium.Duringthistime,allthecellsoftheculturedifferentiateintoalimitednumberofphenotypeswithneuroepithelialandneuronalcellspredominating.Differentiationcouldbeobtainedinthedefinedmediumatrelativelylowcelldensities.Exogenousfibronectinisrequiredforcellattachmenttothesubstratum,andwhenabsentthecellsformaggregatesinwhichdifferentiationstilloccurs.Lowamountsofserumaddedtothedefinedmediumallowmultiplicationandmaintenanceofcellsofundifferentiatedphenotypeandpreventdifferentiationintoneuronalcells.
