Analysis of developmentally homogeneous neural crest cell populations in vitro: II. A tumor-promoter (TPA) delays differentiation and promotes cell proliferation☆

[摘要] Thetumor-promotingagent12-O-tetradecanoylphorbol-13-acetate(TPA)delayedpigmentationindevelopmentallyhomogeneousquailneuralcrestcellpopulationsinvitro.Dosesbelow5×10−8Mresultedonlyinadelayinpigmentationforupto2dayswithoutchangingtherateofpigmentation,whereasintermediatedoses(5×10−8–10−7M)delayedtheinitiationanddecreasedtherateofpigmentationbuthadnoeffectontheultimatefate.DosesofTPAabove2×10−7Mirreversiblypreventedpigmentationinsomeofthecells.TPAdidnotinfluencegrowthoftheundifferentiatedcellpopulation.SinceTPAdelayedthedeclineinthecellproliferationratethatnormallyoccurredwithpigmentation,however,thefinalnumberofpigmentedcellscouldbeincreasedupto10-foldinTPA-treatedcultureswithoutchangingthehomogeneityofthecultures.Apartfromtheeffectsoncelldifferentiation,TPAcausedanimmediatechangeincellularassociationwithintheclusters.
