An essential role for putrescine biosynthesis during meiotic maturation of amphibian oocytes☆
[摘要] TheobjectiveofthisstudywastoinvestigatetheroleofpolyaminesduringmeioticmaturationofXenopusoocytes.Theresultsindicatearapidandsignificantincreaseintheactivityofornithinedecarboxylase(ODC),therate-limitingenzymeinthepolyaminebiosyntheticpathway,duringthemeioticmaturationinducedbyeitherprogesteroneorhumanchorionicgonadotropin(HCG).Thisincreaseintheenzymeactivitywasfollowedbyanaccumulationofputrescinewithoutanyeffectonthelevelsofspermidineorspermine.TheinhibitionofODCactivityandtheaccumulationofputrescinelevelsbyα-difluoromethylornithine(DFMO),acatalyticirreversibleinhibitorofODC,alsoresultedintheinhibitionofmaturationmediatedbyprogesteroneinXenopusoocytes.DFMOcausedaninhibitionofbothmaturationandovulationinducedbyHCGinovarianfragments.Thisinhibitionwasreadilyreversiblebyexogenoussupplyofputrescinetothemedium.Theseobservationssuggestthatputrescineplaysanimportantroleduringthemeioticmaturationofamphibianoocytes.
