Tyrosinase activity in the Wolffian lens regenerating system☆
[摘要] ThefirstpartofthepaperisdevotedtoseparationandcharacterizationoftyrosinaseofNotophthalmusviridescens.AfterammoniumsulfatefractionationandDEAE-cellulosechromatography,afractionofextractwasobtainedwhichhada1000-foldincreaseinenzymeactivityandrevealedonepositiveDOPAoxidasebandandonenegativeproteinbandonacrylamidegelelectrophoresis.Somecharacterizationoftheenzymewasmadeusingthissample.Specificactivityofthisfractionwasestimatedtobe39×10−2μmoleofl-tyrosine/mgin17hr.AninhibitoroftyrosinasewasdemonstratedintheNotophthalmuslens.Inthesecondpartofthepaper,theleveloftyrosinaseactivitywasfollowedintheiristissueasitwastransformedintothelensafterlentectomyinadultN.viridescens.WhentheactivitywasbasedonmicrogramsofDNA,thedorsalirisincreasedslightlyinactivityduringthephaseofdepigmentation.Asignificantincreaseinactivitywasrecordedwhenthelenshadbeenformedbycompletelydepigmenteddorsaliriscells.Ontheotherhand,intheventraliris,whereweakdepigmentationisfollowedbyrepigmentation,theactivitylevelshowedaweakincrease.Implicationofthedatafortissuetransformationwasdiscussed.
