Isolation of a choriolytic enzyme (hatching enzyme) of the teleost, Oryzias latipes☆
[摘要] Achoriolyticenzyme(hatchingenzyme)oftheteleostOryziaslatipeswasisolatedfromtheculturemediuminwhichmanylarvaehatchedoutsynchronouslybysuccessivefractionationwith(NH4)2SO4precipitation,SephadexgelfiltrationandCM-cellulosecolumnchromatography.TheenzymeproteinisfoundtorepresentasinglebanduponstarchgelelectrophoresisatpH8.6and5.2,respectively.Thespecificactivityoftheisolatedenzymepreparationisapproximately180to210timesthatoftheoriginalculturemediumasdeterminedbytheassayofproteolyticactivitytowardcaseinandofclearing(choriolytic)activitytowardnaturalsubstrate,chorion.Themolecularweightoftheenzymeasestimatedroughlybygelfiltrationisapproximately8000.Althoughitseemsprobablethattherearemorethanonechoriolyticand/orproteolyticenzymesintheoriginalmedium,theisolatedenzymeisfoundtohaveapowerfulclearingactivityaswellasproteolyticactivityandtoberesponsibleforaneffectivechoriolysisbyitself.
