Effects of dichlorophenamide, zinc, and manganese on otolith development in mice☆
[摘要] Evidenceispresentedsuggestingthatcarbonicanhydraseandzincaffectthedepositionofotoconia,orcalciumcarbonatecrystals,duringotolithdevelopmentinmice.Asingleinjectionofdichlorophenamide(aninhibitorofcarbonicanhydrase)intopregnantSwiss/COXalbinofemalesataspecificdayofgestation(13–18)producedotolithdefectsintheprogenyasobservedshortlyafterbirth.Similardefectswerefoundinprogenyofpregnantfemalesfedzinc-deficientdietsbeginningatvariousdaysofgestation(0,10–16).Despitethehighlysignificanteffectsofthedrugandofthezinc-deficientdiet,untreatedcontrolsexhibitedalowincidence(approximately15%)ofotolithdefects.Variouspurifieddietswereemployedtomeasuretheeffectsofvaryingratiosofmanganeseandzinconotoliths.Dietsfortifiedwithmanganesepluszinccompletelyeliminatedotolithdefects.Allotherdietsemployed,significantlyreducedtheincidenceofdefects.Itispostulatedthat:(1)themostactiverateofcrystalformationoccursduringdays15and16ofgestation;(2)anobservedasymmetrymayreflectdifferencesintherateofdevelopmentofearprimordia,orinlevelsofcarbonicanhydraseactivity;(3)theotoconiamayundergoarapidmetabolicturnover,ortheymaybeaffectedbysubtlechangesinpHoftheendolymph;and(4)manganeseandzincareessentialfornormalotolithdevelopment.
