Development of mouse limb buds in organ culture: Chondrogenesis in the presence of a proline analog, l-azetidine-2-carboxylic acid☆
[摘要] Limbbudsfrom36–40-somitemouseembryoswereculturedonfluidmediumconsistingofBGJandfetalcalfserum.After6daysthelimbbudsshowedgoodchondrogenesisofrecognizablebonesegments.Inthepresenceoftheprolineanalog,limbl-azetidine-2-carboxylicacid,growthoftheentirelimbbudwasinhibitedascomparedwithcontrolcultures,chondrogenesiswasdelayed,andthecartilagewhichsubsequentlyappearedbecameswollen,soft,andmalformed.Histologicallythematrixofthistreatedcartilageappearedalmostunstained,exceptforstrandsofalcianblue-positivematerialaroundthedisorientedchondrocytes.Growthanddevelopmentofthelimbbudswerenormalwhenadequatel-prolinewasaddedtothemediumalongwiththeanalog,orwhend-azetidinewasusedasthecontrol.l-Azetidine-treatedcultureswhichwereremovedfromtheanalogandsubsequentlyplacedonaproline-richmediumshowedrapidrecoveryofthecartilagetonormalwithin48hr.Thesefindingsagreewellwithpreviousreportsthatl-azetidineinhibitsthesecretionofnormalcollagen:thegrossandmicroscopicchangesobservedreflectthiscollagendeficiencyandemphasizetheimportanceofcollageninnormalorganizationofthematrixandtheresultantshapeofthecartilage.
