Cellular heterogeneity in the chick wing mesoblast: I. Analysis by equilibrium density gradient centrifugation☆
[摘要] Ananalyticandpreparativecellseparationtechnique,equilibriumdensitygradientcentrifugation,hasbeenappliedtotheheterogeneouscellpopulationofthedevelopingchickwingmesoblastinanefforttoisolateseparatelyandcharacterizecellpopulationsdifferinginbuoyantdensity.Whendissociatedcellsuspensionsaredistributedthroughoutacontinuousbovineplasmaalbumin-balancedsaltsolutiongradientandcentrifugedtoequilibrium,reproduciblepatternscharacteristicofthetissuesourceordevelopmentalstageoforiginareobtained.Suchpatternsdifferbothqualitativelyandquantitatively.Morethan90%ofthecentrifugedcellsremainviableasjudgedbydyeexclusion,andnearly90%arerecoveredfromthegradients.Cellularsubpopulationspreparedonthesegradientscanbeclonedyieldingcartilage,muscle,orfibroblasticcolonies.Atstage25thereappearsapopulationofcellswithameanbuoyantcelldensityof1.0437g-cm−3whichexhibitsamarkedlyenhancedabilitytoformcartilage-makingcloneswhencomparedtocellsofdifferingdensityordevelopmentalage.
