Developmental control of α-1–4 glucan phosphorylase in the cellular slime mold Dictyostelium discoideum☆☆☆
[摘要] Glycogenphosphorylaseactivityisundetectableinvegetative,multiplyingcellsofwildtypeandmutantstrainsofDictyosteliumdiscoideumbutappearsduringdevelopment.Thespecificactivityofthisenzymeisundetectableindevelopingcellsuntilmid-aggregationandreachesamaximumatmid-culmination.Thereaftertheenzymeactivitybeginstodecreaseandeventuallydisappears.AppropriateexperimentswithactinomycinDandcycloheximidesuggestthatboththeappearanceanddisappearanceofglycogenphosphorylaserequirepriorRNAandconcomitantproteinsynthesis.TheactivityofthisenzymeincellsofDictyosteliumdiscoideumappearsthereforetobedevelopmentallyregulated.FurtherstudieswithmutantsofD.discoideumsupportthisview.Theenzymewaspurified100-foldfromcrudeextractsanditspropertieswereexamined.Theseresultsarediscussedinthelightofwhatisknownabouttheregulationofpolysaccharidemetabolismduringculmination.
