Human teratogenesis: In vitro studies on thalidomide-inhibited chondrogenesis☆
[摘要] Chondrogenictissuesfrom5to8.5-wk-oldhumanembryoswereexposedinvitrototheteratogenicdrugthalidomide.Theresultssuggestthatamajorsiteofactionofthalidomideisupontissueassociatedwiththemesonephros.Humanmesonephrosfromembryos5–7wkoldcansynthesizeinvitroanextracellularmatrixwhichhasallthecharacteristics,histologicalandbiochemical,ofcartilage.Theabilityofembryonicmesonephrostoformcartilageinvitroisassociated,inanasyetunknownmanner,withinvitrolimbchondrogenesis.Itisproposedthatmesonephricmesenchymeataspecificstageofdevelopmenthasapositiveinfluenceuponlimbchondrogenesis.Thalidomideinhibitsthisinteraction,andinhibitschondrogenesis.Nootherchondrogenictissue,evenisolatedlimbtissue,wasfoundtobeinhibitedbythalidomidetreatment.
