Demonstrability of some oxidative enzymes in early rodent embryos with and without fixation☆
[摘要] Severaloxidativeenzymes[NADH-TR(reducednicotinamide-adeninedinucleotide-tetrazoliumreductase),NADPH-TR(reducednicotinamide-adeninedinucleotidephosphate-tetrazoliumreductase),SDH(succinicdehydrogenase)andLDH(lactatedehydrogenase)]werestudiedbyhistochemicalmeansduringearlydevelopmentofratandmouse.Allinvestigatedenzymescouldbeeasilydemonstratedinzygoteandalsotosomeextentinsomiticstageswithoutanypretreatment.However,incleavageandearlypostimplantationstagesenzymeactivitycouldberevealedonlyaftertheembryoswerepretreatedinsomeway.Thispretreatmentcanbefixationwithformalinoracetone,freezingandthawing,slightmechanicaldamageorveryprolongedincubationtime.TheformazangranulesasasignofenzymaticactivitywerepresentinallstagesofembryonicdevelopmentandweremoreabundantinreactionsforNADH-TRandLDHthaninreactionsforNADPH-TRandSDH.Ourresultssuggestthattheinvestigatedenzymesarepresentinallembryoniccellsduringearlydevelopment.Itseemsthatthepermeabilityofembryoniccellsforhistochemicalmediamustbeincreasedotherwisethehistochemicalreactionscannotbeaccomplished.
