Synthesis of satellite DNA in Rhynchosciara hollaenderi☆
[摘要] DuringthelatterpartofthefourthinstardevelopmentofRhynchosciaralarvae,DNAsynthesisoccursinbothgermcellsandsomaticcells,eventhoughthesecellsdonotundergomitosis.CsCldensitygradientanalysishasrevealedthesynthesisofd(AT)satelliteDNAsinthetestisandinsomatictissuessuchassalivarygland,fatbody,andgastriccecum.Inthesestudiesithasbeenshownthatthereisatissue-specificvariationintherelativeproportionsofsynthesisofd(AT)satelliteandmain-bandDNAinthetestisduringthefourthinstar.Theinitiationofsynthesisofthed(AT)satelliteinthetestiscorrespondswiththeappearanceandformationofthehighlycharacteristicmitochondriawhichdevelopduringthematurationofthespermatocytes.ThissatelliteDNAhasbeenshowntobecircularandhasadensityof1.681gm/cm3inCsCl.SynthesisofasimilarcircularDNAcannotbedetectedinsomatictissues,althoughthesetissuesdosynthesizead(AT)satelliteofsimilardensity.WeconcludefromthesestudiesthatthespecificsynthesisofauniquecircularDNAmaybeanessentialpartofthemorphogeneticdevelopmentofthehighlyspecializedmitochondriaformedduringspermatocytematuration.
