[摘要] ThepresenceofglycogeninArbaciapunctulataembryoswasdeterminedafteralkalineextractionbyaspecificassayutilizingcrystallineglycogenphosphorylaseandamylo-1,6-glucosidase.Matureunfertilizedeggscontainanaverage23.4μgofglycogenpermilligramofprotein.Glycogenreservesaremaintainedafterfertilizationthroughtheblastulastagethusarenotutilizedastheprimaryenergysourceforearlydevelopment.Atgastrulationthereisadroptoanaverage14.8μgofglycogenpermilligramofprotein.Thepolysaccharidemaycontributesignificantlytotheenergypoolatthisstage.Glycogenwasisolatedfrommatureovaandpreparedforelectronmicroscopy.Thenegativelystainedmoleculesaresingleparticles300–400Ã…indiameter.Glycogenpoolsarenotinertduringdevelopment.GrowingembryoswereculturedinthepresenceNaH14CO3.Incorporationofisotopeintoglycogenwasobservedfromthe2-cellstageonward.Byblastula,glycogen-14Caccountedfor18%ofthetotalradioactivityincorporated.Glycogenphosphorylase(pHoptimum7.3)ispresentinunfertilizedeggsandmorulae.Theenzymeinbothinvitrosystemsiscapableofgettingridofalltheegg'sglycogenwithin1hr.LackofexogenousAMPreducesphosphorylaseactivityby50%inenzymepreparationsofunfertilizedeggs.
