Some effects of trypsin dissociation on the inhibition of reaggregation among embryonic chicken neural retina cells by cycloheximide☆
[摘要] Theeffectsofcycloheximideonthereaggregationoftrypsin-dispersed,embryonicchicken,neuralretinacellswereinvestigated.Thecellswereusedeitherimmediatelyafterisolation(F-cells),orafter24hrpriorcultureunderconditionsnotpermittingreaggregation(V-cells).Theparametersofaggregationusedwerethesizeofaggregatesformedingyrotatoryshakercultures,andtheconcentrationofsinglecellsinthesecultures.Atboth24and48hr,followingtreatmentwithcycloheximide,themeandiametersoftheaggregatesofF-cellsshowedagreaterreductionthantheV-cells,whencomparedwiththeircorrespondingcontrols.Inaddition,cycloheximideresultedinahigherconcentrationofsinglecells,thatislesscell/celladhesion,thanincomparablecontrols.AhigherproportionofsinglecellswerepresentintheF-cellculturesinthepresenceofcycloheximidethanintheV-cellcultures.Thus,thedegreeofinhibitionofcelladhesionbyoneinhibitorofproteinsynthesisdifferedinF-cellsandV-cells.Theseexperimentsmayservetofocusattentiononsomesecondaryeffectsoftrypticdissociation,whichisanoftenoverlookedfactorinsubsequentstudiesofreaggregatingembryoniccells,particularlyinthoseexperimentsinvolvingtheuseofmetabolicinhibitors.
