Cellular energy metabolism during fetal development: IV. Fatty acid activation, acyl transfer and fatty acid oxidation during development of the chick and rat☆
[摘要] WehaveinvestigateddevelopmentalprofilesofATP-dependentpalmityl-CoAsynthetase,acetyl-CoAsynthetase,palmitylcarnitinetransferase,andfattyacidoxidationinheartandliverofdevelopingchicksandrats.Palmityl-CoAsynthetaseactivityofratliverandhearthomogenatesincreased6-to10-foldduringthefirstpostnatalweek.Chickembryoheartactivitypeakedbetween13and16daysofdevelopment.Theactivityofembryonicchickliverswasbimodalwithhighestactivityseenat7and16daysofdevelopment.Posthatchingvalueswereapproximately50–75%ofthepeakembryoniclevels.Acetyl-CoAsynthetaseactivityofratliverandhearthomogenateswaslowbutalsoshoweddevelopmentalincreasesfollowingbirth.Acetyl-CoAsynthetaseactivityofchickembryonicheartswasgreatestat16daysofdevelopment.Palmitylcarnitinetransferaseactivityofratliverandhearthomogenatesshowedastrikingincreaseduringthefirstweekoflife.Chickheartactivitywassimilartothatobservedforpalmityl-CoAsynthetasewithapeakbetween13and16daysofembryonicdevelopment.Coincidentwiththepostnatalriseinfattyacidactivationandpalmitylcarnitinetransferaseactivityindevelopingrats,theoxidationofpalmityl-CoApluscarnitineandofpalmitylcarnitineincreasedfrombarelymeasurablelevelsatbirthtoadultlevelsby30daysofage.Theincreasesthatweobserveprobablyrelatetochangesinthespecificactivityoftheenzymesaswellastoanincreaseintheabsolutenumberofmitochondriaduringdevelopment.
