Pyridine nucleotides in normal and nicotinamide depleted adrenal tumor cell cultures☆
[摘要] MouseadrenaltumorcelllineY-129wasgrownincellcultureinmediumwithoutnicotinamide.Inhibitionofgrowthoccurredafterthesecondsubcultureinthevitamin-deficientmedium.Pyridinenucleotidesweremeasuredincontrolandnicotinamide-starvedcultures.DPN+decreasedtolessthan10%ofthenormallevelafter7daysofvitaminstarvation.TPNHdroppedto25%ofitsnormallevelinthesameperiod.Platingefficiencydecreasedfrom20%incontrolsto10%in7-daynicotinamide-starvedcultures.Steroidproductionwasreducedbyapproximately50%inthestarvedcultures.Sensitivityoftheculturestothelethaleffectsofthenicotinamideantagonist,3-acetylpyridine,wasinverselyproportionaltothepyridinenucleotidecontentofthecells.Reconstitutionofnicotinamideindepletedculturescausedarapidrenewalofthepyridinenucleotidelevelsandcompleteprotectionagainsttheeffectsof3-acetylpyridine.Recoveryandsubsequentgrowthoftheculturesdidnotrestorethedecreasedcapacitytoproducesteroids.NicotinicacidwasnotutilizedbythecellsforthesynthesisofpyridinenucleotidesindepletedculturesandazaserinedidnotinhibittheutilizationofnicotinamideindicatingthatDPN+wassynthesizeddirectlyvianicotinamideribonucleotide.
