Properties of electrotonic junctions between embryonic cells of Fundulus☆
[摘要] CellsisolatedfromblastulaeoftheteleostFunduluscanbereaggregatedinpairsinphysiologicalsaltsolution.Thecellpairsusuallybecomeelectricallycoupled.Unlikemanycoupledcellsofadulttissues,injectedfluoresceindoesnotpassbetweenthesecellsindetectableamounts,althoughitcanenterthecellswhenexternallyapplied.Glutaraldehydefixationrapidlyandgreatlyincreasescouplingresistance,butcausesonlymoderateincreasesincellresistanceexclusiveofthecouplingpathway;thusfixationdecouplescells.Electricalmeasurementsdemonstratethatfixationdoesnotdisruptzonulaeoccludentesinblastulaeorgastrulae.ElectronmicroscopyrevealsthattheappositionalregionbetweenreaggregatedcellpairsispenetratedbyLa(OH)3appliedwiththefixative.Therearealsoextensiveregionsofcloseappositionbetweenthecellmembranes.Thecombineddataindicatethattheseembryoniccellsarecoupledbywayofspecialized,electrotonicjunctionsratherthanaregionofextracellularspacebetweenthemthatiswalledoffbyazonulaoccludens.Amorphologicalsubstrateforthesejunctionsisprovidedbythecloseappositionsseenelectronmicroscopically.Therelativelyrestrictivepermeabilityoftheelectrotonicjunctionsbetweenembryoniccellsmayallowsmallionstofunctionasintercellularsignalingmoleculeswhilerestrictingmovementoflargermoleculesinvolvedinintracellularcontrols.
