Thallium-induced achondroplasia in the embryonic chick
[摘要] Achondroplasiahasbeeninducedintheembryonicchickbytheadministrationof0.6mgofthalliumsulfateontothechorioallantoicmembraneat7daysofincubation.Embryoswerestudiedat2-dayintervalsto18daysofincubation,andthelongboneswereexaminedhistologically,histochemically(collagen,acidmucopolysaccharide,glycogen,calcification,alkalinephosphatase)andbiochemically(hydroxyprolineandhexosamineanalyses).Growthofthelongboneswasreducedandareasofnecroticcartilageformedinthetreatedembryos.Thenecroticchondrocytesaccumulatedcollagen,calcium,andlipidandcontainedlessthannormalamountsofacidmucopolysaccharide.Totalacidmucopolysaccharidewasreducedinthelongbonesfromtreatedembryosbecauseofdefectivesynthesisand/orreleasewithinthecartilaginousfraction.Thalliumproducesitseffectbythisinhibitionofacidmucopolysaccharideandthismechanismhasbeendiscussedinrelationtoachondroplasiaofgeneticorigin.
