Dedifferentiation of iris epithelial cells☆
[摘要] Electronandphase-contrastmicroscopicobservationsdemonstratedthatdepigmentationofirisepithelialcellsinvivoafterlentectomyisprecededbyalterationsofcellshapeandincreasesinmicrofilamentsandmicrotubulesintheperipheryofthecell.Extensivecellprocessesareformed,withtipsbranchingintofinestrandsthatcontainmelanosomes.Themacrophagesinvadingtheirisepitheliumincorporatepiecesofsuchstrands,whicharecomposedofcellmembrane,cytoplasmicmatrix,andmelanosomes.Individualmelanosomesarealsotakenupbymacrophages.Someofthestrandsofiriscellprocessesseemtodegeneratewithintheintercellularspace.Thus,depigmentationofirisepithelialcellsisaccompaniedbylossofasubstantialpartofthecellsurfaceandcytoplasmicmatrixofirisepithelialcells.Measurementsoftheabsolutevolumeofwholeirisepithelialcells,theircytoplasm,andtheirnucleiwereconductedatvariousstagesofdepigmentation.Thesemeasurementsrevealanextensiveincreaseinthevolumeofthecytoplasmprecedingactivationofthecellsurfaceandasignificantreductionincytoplasmicvolumeduringthephaseofextensivedepigmentation.Inthecaseofthenuclearvolume,theextensiveincreasewhichoccursinparallelwiththatofthecytoplasmicvolumeisnotfollowedbyasignificantchangeduringthedepigmentationphase.
