Kinetic analysis of amino acid pools and protein synthesis in amphibian oocytes and embryos☆☆☆
[摘要] RatesofproteinsynthesishavebeenmeasuredinRanapipiensoocytesandembryosandinXenopusoocytesfromtheincorporationkineticsoftwodifferentconcentrationsofaminoacid.Thismethoddoesnotrequireanindependentmeasurementoftheaminoacidpools,sincethepoolsizecanbecalculateddirectlyfromincorporationdata.Theeffectsoftheconcentrationanddiffusionofinjectedaminoacidonthecalculatedvaluesforaminoacidpoolsizeandflowratearediscussed.Whentheendogenousaminoacidpoolisappreciablyexpandedbytheinjectedaminoacid,thetotalaminoacidpoolintheoocytesorembryosmaybeconsideredastheprecursorpoolforproteinsynthesis.Underthesecircumstances,compartmentationofaminoacidsdoesnotaffecttheresults,exceptwhenlysineisusedastracer.TheratesofproteinsynthesisinovarianoocytesofRanapipiensandXenopuslaevisare18and50–54ng/hr,respectively.InRanapipiens,therateincreases70%duringmaturationandanother50%beforethetwo-cellstage.Finally,therateapproximatelydoublesbetweenthetwo-cellandblastulastages.
